What is Gonadorelin 2 mg?

Gonadorelin (gonadorelin acetate) is a synthetic form of natural GnRH — gonadotropin-releasing hormone (a decapeptide). As a GnRH receptor agonist it stimulates the pituitary gland to secrete LH (luteinizing hormone) and FSH (follicle-stimulating hormone), which in turn stimulate the gonads. In the research literature it is used for research on the hypothalamic–pituitary–gonadal (HPG/HPT) axis, fertility and hormonal balance.

Gonadorelin belongs to the category peptides for immunity and hormonal balance. It acts on the HPG axis at the pituitary level — between the „higher"-positioned Kisspeptin-10 (which stimulates the secretion of the body's own GnRH) and the „lower"-positioned HCG (which mimics LH itself).

It is a research compound — it is not a medicinal product and holds no registration for clinical use in the EU in this form. (Gonadorelin is known in reproductive endocrinology, but the product offered by Peptology is a research-grade reagent, not intended for diagnosis or treatment.)

In the version offered by Peptology, Gonadorelin is available in a 2 mg vial as a lyophilized peptide of high purity, confirmed by independent laboratory analysis (CoA for every batch). The product is intended exclusively for scientific research and in vitro applications.

Variant: Gonadorelin 2 mg

This page concerns a single dose variant — Gonadorelin 2 mg. Each vial contains 2 mg of lyophilized peptide. This is a sufficient amount for research protocols with pulsatile dosing (characteristic of GnRH), where very small portions are used (e.g. in the microgram range).

REFERENCE CONCENTRATION

2 mg/mL or 1 mg/mL

2 mg in 1 mL = 2 mg/mL; in 2 mL = 1 mg/mL. Calculate in the calculator.

Gonadorelin is offered in the 2 mg variant. Other HPG-axis compounds are Kisspeptin-10 (a level higher) and HCG (a level lower, mimics LH).

How Gonadorelin works — the HPG axis

In the research literature, Gonadorelin is described as a GnRH agonist acting on the pituitary. Below are the three most frequently studied elements.

GnRH RECEPTOR

Binds the GnRH receptors in the pituitary and stimulates the release of LH and FSH — it mimics physiological GnRH.

LH / FSH → GONADS

LH and FSH stimulate the gonads → production of testosterone/estrogens and spermatogenesis.

PULSATILE ADMINISTRATION

The action depends on the administration mode: pulsatile supports the axis, continuous may inhibit it (desensitization).

Why the pulsatile mode is key

Physiologically, GnRH is secreted pulsatilely, not continuously. That is why in research protocols Gonadorelin is administered in „pulses" (e.g. a described schedule of 5 µg every 90 min) — this rhythm supports the HPG axis. Continuous administration can paradoxically inhibit it (receptor desensitization) — this is a mechanism used for a purpose other than stimulation.

Gonadorelin acts „one level higher" than HCG — it stimulates the natural pathway through the pituitary (LH + FSH), while HCG mimics LH itself, bypassing the pituitary. These data come from research on the HPG axis and do not constitute a claim about the effect of the research-grade product offered by Peptology.

Pharmacokinetics and pharmacodynamics (research framework)

Gonadorelin is a short-acting decapeptide with a rapid but transient effect on gonadotropins. The most important research features:

  • Class: synthetic GnRH (decapeptide), GnRH receptor agonist
  • Target: GnRH receptors in the pituitary → release of LH and FSH
  • Onset of action: an LH/FSH increase observed within ~30–90 min of administration
  • Administration mode: pulsatile (supports the axis) vs continuous (desensitization) — a key distinction
  • Short duration of action: a rapid and transient effect, hence the pulsatile schedules
  • Route of administration in protocols: subcutaneous or by infusion pump (pulsatilely), per the protocol

Comparison: Gonadorelin vs Kisspeptin-10 vs HCG

Three compounds from the same category act on the HPG axis at different levels. Kisspeptin-10 is „higher" (stimulates GnRH), Gonadorelin acts on the pituitary (GnRH → LH/FSH), and HCG is „lower" (mimics LH).

Three levels of the same axis: Kisspeptin-10 stimulates GnRH secretion, Gonadorelin acts like GnRH on the pituitary (LH+FSH), and HCG mimics LH itself, bypassing the pituitary. This comparison is indicative in nature and does not constitute a recommendation for use.

Research areas for Gonadorelin

Gonadorelin is a classic tool of reproductive endocrinology. Below are the most frequently explored research directions.

LH/FSH stimulation and pituitary diagnostics

Gonadorelin is studied as a tool for assessing pituitary function — it produces a marked increase in LH and FSH within 30–90 minutes, which is used in HPG-axis diagnostic models.

Fertility and the hormonal axis

In fertility research, Gonadorelin (in pulsatile mode) is used to support the HPG axis — including in models of amenorrhea (a described schedule of 5 µg every 90 min). It is sometimes considered together with Kisspeptin, HMG or clomiphene in research protocols.

Axis restoration (PCT)

Gonadorelin is often considered in protocols for restoring the HPG axis after its suppression (e.g. after an androgen cycle, PCT), where the goal is to resume natural production of LH/FSH and sex hormones. These are research directions, not confirmed applications of the research-grade product.

Research dosing protocols

Important: The information below describes schedules used in research on Gonadorelin. It is provided for scientific and informational purposes and does not constitute a recommendation for individual use, medical advice or administration instructions. The research-grade Gonadorelin offered by Peptology is not a registered medicine.

In research protocols, Gonadorelin is usually administered after reconstitution of the lyophilizate, most often subcutaneously and — crucially — pulsatilely:

  • The pulsatile mode (e.g. a described schedule of 5 µg every 90 min) supports the HPG axis; continuous administration can inhibit it (desensitization)
  • Very small unit doses (micrograms) — a 2 mg vial is enough for many portions
  • The effect on LH/FSH observed within ~30–90 min of administration
  • Calculate precise volumes and units in the peptide calculator (max. 3 mL of solvent per vial)

In combination protocols, Gonadorelin is sometimes considered together with Kisspeptin-10 or other compounds of the reproductive axis. These schedules come from the research literature and do not constitute a recommendation for self-administration.

Side effects and safety

In research observations, Gonadorelin is described as well tolerated. Side effects are usually mild, transient and reversible. The data concern research on the molecule.

Common (mild)

  • Headache or mild dizziness
  • Hot flushes / redness
  • Local irritation after injection

Tolerability profile

  • Well tolerated in observations
  • Symptoms transient and reversible
  • No toxicity at the described doses

To consider

  • Slight increase in heart rate
  • Continuous mode may inhibit the axis (desensitization)
  • Hypersensitivity to components

Most reported symptoms are mild and reversible. The distinction of administration mode is crucial — pulsatile supports the axis, continuous may inhibit it. This information is scientific in nature and does not constitute an instruction for use.

Storage and reconstitution

Gonadorelin is supplied as a lyophilized (freeze-dried) powder in a vial. In lyophilized form it is stable at room temperature, protected from light and moisture — refrigeration before reconstitution is not mandatory. After reconstitution, stability depends on the solvent used, with Peptide Matrix Water™ significantly extending the peptide's lifespan.

  • Before reconstitution: room temperature, in the original packaging, protected from light and moisture. A refrigerator is not mandatory.
  • After reconstitution with Peptide Matrix Water™: stability in the refrigerator +2 °C to +8 °C for up to 60 days under appropriate laboratory conditions — the stabilizing formula extends the peptide's lifespan relative to standard solvents.
  • After reconstitution with bacteriostatic or sterile water: stability in the refrigerator +2 °C to +8 °C for up to 28 days under appropriate conditions. Do not refreeze the reconstituted solution.
  • Reconstitution: in a laboratory setting, Peptide Matrix Water™, bacteriostatic water or sterile water for injection is usually used, depending on the protocol.
  • Volume: maximum 3 mL of solvent (capacity of a standard vial). For a precise calculation of volume and units, use the peptide calculator.
  • Homogenization: add the solvent slowly down the vial wall, without vigorous shaking, then wait 10–15 min. Swirl gently, do not shake.

Laboratory tip: With your order, you can add Peptide Matrix Water™ to the cart — a stabilizing solution designed for working with reconstituted research peptides. More in the article Bacteriostatic water vs acetic acid vs matrix water.

Frequently asked questions about Gonadorelin

What is the difference between Gonadorelin and HCG?

Gonadorelin stimulates the pituitary to release LH and FSH (it acts „higher", supporting the natural pathway), whereas HCG mimics LH itself, bypassing the pituitary. These are two different levels of the same HPG axis.

And how does it differ from Kisspeptin-10?

Kisspeptin-10 acts „one level higher" — it stimulates the hypothalamus to secrete its own GnRH, while Gonadorelin is itself GnRH acting on the pituitary. Kisspeptin thus works with the natural rhythm, and Gonadorelin delivers the GnRH signal directly.

Why is pulsatile administration important?

GnRH is physiologically secreted in pulses. Pulsatile administration of Gonadorelin supports the HPG axis, whereas continuous administration can paradoxically inhibit it (receptor desensitization). The mode of administration determines the effect.

Is it suitable for PCT?

In the research literature, Gonadorelin is sometimes considered in protocols for restoring the HPG axis after its suppression (PCT). This is, however, a research context, not a usage recommendation — the research-grade product is not a medicine.

How is Gonadorelin stored and reconstituted?

The lyophilized vial is stored at room temperature, protected from light and moisture. After reconstitution, stability depends on the solvent: with Peptide Matrix Water™ — up to 60 days in the refrigerator +2 °C to +8 °C; with bacteriostatic or sterile water — up to 28 days. Avoid refreezing.

What you need

Precision pipette

Calibrated, 100–1000 µL — for accurately measuring the volume of solvent added to the vial.

70% alcohol swabs

Two swabs — to disinfect the peptide vial and the solvent vial.

Peptide Matrix Water

Stabilized solvent for lyophilized peptides. Why this one.

Five rules for reconstitution

✅ Correct ❌ Mistake
Disinfect the vial caps and the work surface with 70% alcohol; wear gloves Working without disinfection
Add the solvent slowly along the vial wall Direct jet onto the powder — foaming and denaturation
Gentle rolling between the palms Shaking — foam, which denatures peptides
10–15 minutes waiting at room temperature Mechanical mixing or heating
Visual check: clear, particle-free Working with a cloudy or discolored solution

Calculating concentration

⚠️ Maximum 3 mL solvent. The standard vial does not accommodate larger volumes — do not exceed during reconstitution.

  • Concentration (mg/mL) = mg in the vial ÷ mL of solvent
  • Volume per sample (mL) = amount per sample (mg) ÷ concentration (mg/mL)
  • 1 mg/mL = 1 µg/µL · 1 mL = 1000 µL

Reconstitution per vial — solvent: Peptide Matrix Water. Concentration after adding the solvent (1 mg/mL = 1 µg/µL):

Vial+ 1 mL+ 2 mL+ 3 mL
2 mg2 mg/mL1 mg/mL0.67 mg/mL

👉 For any vial and solvent volume — use the calculator. It calculates the concentration (mg/mL, µg/µL) and the volume per sample (mL, µL).

Storage

BEFORE RECONSTITUTION

Room temperature, protected from direct light, in original packaging. Refrigerator not required.

AFTER RECONSTITUTION

+2 °C to +8 °C, up to 60 days with Peptide Matrix Water. Discard any remainder after this period.

FREEZING

Not recommended. Freeze/thaw cycles degrade peptide structure.

Four mistakes to avoid

1. Shaking "to dissolve faster"

Foam = denaturation. Only gentle rolling between the palms.

2. Inappropriate solvent

For peptide stability use only sterile, specially formulated solvent — recommended Peptide Matrix Water.

3. Ignoring slight turbidity

Not fully clear = the sample does not meet the criteria for use.

4. Taking it out of the refrigerator "for an hour–two"

Short exposures add up and compromise stability.

Research status: All Peptology products are intended for laboratory and research use only. They are not medicinal products, not approved for human or veterinary use and do not substitute medical advice. The described protocol is standard laboratory practice for working with lyophilized peptides.
