What is TB-500 (Thymosin Beta-4)?

TB-500 is a synthetic peptide corresponding to the active fragment of thymosin beta-4 (Tβ4) — a natural, 43-amino-acid protein that the body releases at the site of injury. TB-500 contains the key, actin-binding region of Tβ4, responsible for cell migration, tissue repair and anti-inflammatory action. In the research literature it is described as one of the most important regenerative peptides, targeted at healing and rebuilding soft tissues.

TB-500 belongs to a different class than the GH-axis peptides. Molecules such as CJC-1295 or Ipamorelin stimulate the secretion of growth hormone, and IGF-1 LR3 drives hypertrophy. TB-500 acts differently — it regulates actin and directs repair cells to the damaged tissue, supporting angiogenesis and limiting inflammation. That is why in studies it is often combined with BPC-157 as a complementary regenerative pair.

TB-500 is studied in the context of wound healing, repair of tendons, ligaments and muscles, regeneration of cardiac tissue and modulation of inflammation. It is a research peptide — it holds no authorization as a medicinal product. See also the other recovery and anabolic peptides in our category.

In the version offered by Peptology, TB-500 is supplied as a lyophilized peptide with purity above 99% (HPLC/MS), confirmed by independent laboratory analysis (CoA for every batch). It is available in two vial formats — 5 mg and 10 mg — depending on the scale of the research protocol.

Available formats: 5 mg and 10 mg

TB-500 is available in two lyophilized formats that cover different scales of research work — from single in vitro series to longer regenerative protocols with higher peptide consumption.

5 MG

Standard format

Suitable for validation experiments, single in vitro series and shorter regenerative protocols.

10 MG

Enlarged format

For longer research series, tissue-repair models and protocols with higher peptide consumption.

Both formats have identical purity > 99% and the same laboratory certificate for the batch. The choice of size depends on the scale of the protocol, not on the concentration of the active substance.

How TB-500 works — actin regulation and cell migration

In the research literature, TB-500 (thymosin beta-4) is described as a molecule acting through several complementary repair mechanisms. Their common denominator is accelerating the movement of repair cells to the damaged tissue and rebuilding its blood supply.

ACTIN & MIGRATION

Sequesters G-actin, creating a reservoir for the F-actin network — this drives the migration of fibroblasts, endothelial cells and progenitor cells to the site of injury.

ANGIOGENESIS

Supports the formation of new blood vessels — endothelial cell migration and the formation of vascular structures, improving the blood supply of the healing tissue.

ANTI-INFLAMMATORY & ANTIFIBROTIC

Reduces the production of inflammatory cytokines and apoptosis and limits the formation of myofibroblasts — less scarring, more functional repair.

Why the actin mechanism matters

Most tissue regeneration begins with one limiting step — the repair cells must physically reach the site of damage. Thymosin beta-4 regulates actin, the cytoskeleton responsible for cell movement. Thanks to this, TB-500 is associated in studies with faster cell migration, mobilization of stem and progenitor cells, and better blood supply — which together accelerate healing and limit scar formation.

This mechanism is complementary to BPC-157, which acts mainly cytoprotectively and pro-angiogenically — which is why the two peptides are often studied together. This data comes from preclinical studies and does not constitute a claim about the effect of the research-grade product.

Pharmacokinetics and pharmacodynamics of TB-500

TB-500 is a small, well-soluble peptide, which in the published data is described as a molecule with a relatively short plasma half-life but with effects that last longer thanks to redistribution to tissues.

  • Plasma half-life: on the order of 2–3 hours (data from animal models after SC/IM administration)
  • Distribution: low molecular weight favors good distribution to tissues and redistribution after the plasma phase
  • Profile of action: tissue effects last longer than the presence of the peptide in plasma
  • Target: actin regulation (G-actin sequestration), migration and repair signaling
  • Route of administration in protocols: subcutaneous or intramuscular, per the research protocol
  • Schedules: loading and maintenance phases are described in the literature, often in cycles

TB-500 vs BPC-157 vs IGF-1 LR3

The table below compares the three most frequently studied "regenerative" peptides with different mechanisms. TB-500 regulates actin and cell migration, BPC-157 acts cytoprotectively and pro-angiogenically, and IGF-1 LR3 drives growth and hypertrophy. Because their mechanisms are complementary, in studies they are sometimes combined into a "regenerative stack". The data are scientific and comparative in nature.

Research and clinical data on TB-500 / thymosin beta-4

Most data concern the full thymosin beta-4 (Tβ4) molecule; the TB-500 fragment itself is studied mainly preclinically, but shares the same active region with Tβ4. Below are the most important observations from the literature.

Wound healing — Tβ4's flagship area

Wound healing is the best-documented application of thymosin beta-4 and the only one with data from clinical trials in humans. Numerous independent laboratories reproduced the effects of Tβ4 in various injury models — from skin wounds to soft tissues — associating them with faster cell migration and better blood supply.

Clinical trials of full Tβ4 (RGN-259, RGN-137)

Full thymosin beta-4 was the subject of clinical trials (including phase 2) under the codes RGN-259 (dry eye) and RGN-137 (chronic wounds), as well as in neurorestorative programs. These studies do not concern the TB-500 fragment directly, but share its active region and provide context regarding safety and efficacy.

Soft-tissue and cardiac models

In preclinical models, Tβ4 / TB-500 is studied in the context of the repair of tendons, ligaments and muscles and the regeneration of cardiac tissue after ischemia. The results are associated with the mobilization of progenitor cells, angiogenesis and the limitation of fibrosis.

Important: The information below describes schedules used in research on TB-500 / thymosin beta-4. It is provided for scientific and informational purposes and does not constitute a recommendation for individual use, medical advice or administration instructions. The research-grade TB-500 offered by Peptology is not a registered drug.

In the research literature, TB-500 is described in schedules with a loading and maintenance phase, administered subcutaneously or intramuscularly:

  • Loading phase: more frequent administration in the first weeks of the protocol
  • Maintenance phase: less frequent administration sustaining the regenerative effect
  • Subcutaneous or intramuscular administration, per the research protocol
  • Cyclicity: the literature describes cycles with break periods and monitoring of the response

In regeneration protocols, TB-500 is sometimes combined with BPC-157 (complementary mechanisms) and with GH-axis peptides, like CJC-1295 or IGF-1 LR3. These schedules come from the research literature and do not constitute a recommendation for self-administration.

Side effects and safety

In studies and observations, TB-500 is described as well tolerated, with rare and mild side effects. Most of them are transient and dose-dependent.

Frequently observed

  • Mild discomfort at the injection site
  • Transient fatigue or drowsiness (first days)
  • Slight headache
  • A "rush" / warmth sensation after administration

Less frequently described

  • Slightly increased appetite
  • Transient dizziness
  • Local redness

To consider

  • Pro-angiogenic action — caution in proliferation contexts
  • Hypersensitivity to formula components
  • No long-term safety data

Most observed side effects are mild and reversible. As a pro-angiogenic peptide, TB-500 requires caution in contexts related to cell proliferation. This information is scientific in nature and does not constitute an instruction for use.

Storage and reconstitution

TB-500 is supplied as a lyophilized (freeze-dried) powder in a vial. In lyophilized form the peptide is stable at room temperature, protected from light and moisture — refrigeration before reconstitution is not mandatory. After reconstitution, stability depends on the solvent used, with Peptide Matrix Water™ significantly extending the peptide's lifespan.

  • Before reconstitution: room temperature, in the original packaging, protected from light and moisture. A refrigerator is not mandatory.
  • After reconstitution with Peptide Matrix Water™: stability in the refrigerator +2 °C to +8 °C for up to 60 days under appropriate laboratory conditions — the stabilizing formula extends the peptide's lifespan relative to standard solvents.
  • After reconstitution with bacteriostatic or sterile water: stability in the refrigerator +2 °C to +8 °C for up to 28 days under appropriate conditions. Do not refreeze the reconstituted solution.
  • Reconstitution: in a laboratory setting, Peptide Matrix Water™, bacteriostatic water or sterile water for injection is usually used, depending on the protocol.
  • Volume: maximum 3 mL of solvent (capacity of a standard vial). For a precise calculation of volume and units, use the peptide calculator.
  • Homogenization: add the solvent slowly down the vial wall, without vigorous shaking — peptide bonds are sensitive to mechanical stress. Swirl gently, do not shake.

Frequently asked questions about TB-500

What exactly is TB-500?

It is a synthetic peptide corresponding to the active, actin-binding region of thymosin beta-4 — a protein released by the body after injury. It supports cell migration, angiogenesis and anti-inflammatory action, accelerating tissue repair. The results come from the research literature.

How does TB-500 differ from BPC-157?

Both are regenerative peptides, but they act differently. TB-500 regulates actin and cell migration (the movement of repair cells to the injury), while BPC-157 acts mainly cytoprotectively and pro-angiogenically. Their mechanisms are complementary, which is why they are often combined into a pair in studies.

Is TB-500 the same as thymosin beta-4?

TB-500 is a synthetic peptide corresponding to the active fragment of thymosin beta-4 (Tβ4). It shares the same actin-binding region with full Tβ4, which is why data from studies on Tβ4 are an important context, although they do not concern the TB-500 fragment directly.

Can it be combined with other peptides?

Yes — in studies it is classically combined with BPC-157 for a complementary regenerative effect, and also with GH-axis peptides (CJC-1295, Ipamorelin) or with IGF-1 LR3.

Which format to choose — 5 or 10 mg?

Both have identical purity > 99% and the same batch certificate. 5 mg works well for shorter protocols and single series, and 10 mg — for longer tissue-repair models with higher peptide consumption.

How is TB-500 stored and reconstituted?

The lyophilized vial is stored at room temperature, protected from light and moisture. After reconstitution, stability depends on the solvent: with Peptide Matrix Water™ — up to 60 days in the refrigerator +2 °C to +8 °C; with bacteriostatic or sterile water — up to 28 days. Avoid refreezing.

What you need

Precision pipette

Calibrated, 100–1000 µL — for accurately measuring the volume of solvent added to the vial.

70% alcohol swabs

Two swabs — to disinfect the peptide vial and the solvent vial.

Peptide Matrix Water

Stabilized solvent for lyophilized peptides. Why this one.

Five rules for reconstitution

✅ Correct ❌ Mistake
Disinfect the vial caps and the work surface with 70% alcohol; wear gloves Working without disinfection
Add the solvent slowly along the vial wall Direct jet onto the powder — foaming and denaturation
Gentle rolling between the palms Shaking — foam, which denatures peptides
10–15 minutes waiting at room temperature Mechanical mixing or heating
Visual check: clear, particle-free Working with a cloudy or discolored solution

Calculating concentration

⚠️ Maximum 3 mL solvent. The standard vial does not accommodate larger volumes — do not exceed during reconstitution.

  • Concentration (mg/mL) = mg in the vial ÷ mL of solvent
  • Volume per sample (mL) = amount per sample (mg) ÷ concentration (mg/mL)
  • 1 mg/mL = 1 µg/µL · 1 mL = 1000 µL

Reconstitution per vial — solvent: Peptide Matrix Water. Concentration after adding the solvent (1 mg/mL = 1 µg/µL):

Vial+ 1 mL+ 2 mL+ 3 mL
5 mg5 mg/mL2.5 mg/mL1.67 mg/mL
10 mg10 mg/mL5 mg/mL3.33 mg/mL

👉 For any vial and solvent volume — use the calculator. It calculates the concentration (mg/mL, µg/µL) and the volume per sample (mL, µL).

Storage

BEFORE RECONSTITUTION

Room temperature, protected from direct light, in original packaging. Refrigerator not required.

AFTER RECONSTITUTION

+2 °C to +8 °C, up to 60 days with Peptide Matrix Water. Discard any remainder after this period.

FREEZING

Not recommended. Freeze/thaw cycles degrade peptide structure.

Four mistakes to avoid

1. Shaking "to dissolve faster"

Foam = denaturation. Only gentle rolling between the palms.

2. Inappropriate solvent

For peptide stability use only sterile, specially formulated solvent — recommended Peptide Matrix Water.

3. Ignoring slight turbidity

Not fully clear = the sample does not meet the criteria for use.

4. Taking it out of the refrigerator "for an hour–two"

Short exposures add up and compromise stability.
