Retatrutide (Retatrutide) (CAS 2381089-83-2) is an experimental peptide agonist that activates triple GLP-1, GIP and glucagon receptors. It is being studied in clinical trials in obesity, type 2 diabetes and MASLD.
Its structure is designed to include a lipophilic side chain that extends the half-life and allows once-weekly administration in clinical trials.

GLP-1 receptor (GLP-1R): enhances glucose-dependent insulin secretion, delays gastric emptying and suppresses appetite.
GIP receptor (GIPR): Retatrutide is ~8.9 times more potent than native GIP — improves postprandial insulin response.
GCGR (glucagon receptor): although with lower activity (≈30–40%), GCGR activation improves hepatic metabolism, drives lipid mobilization and increases energy expenditure.
Multi-target effect: the combination promotes weight loss through reduced appetite + higher metabolism, as well as improved glycaemic and lipid parameters.

High doses (8 and 12 mg) produced a mean weight loss of 22.8% and 24.2% versus baseline.
Liver fat (LF): at 12 mg — up to a −82.4% reduction after 24 weeks, and −86.0% after 48 weeks.
Mean weight loss: 17.5% at the 12 mg dose.
Lancet study (phase 2): Retatrutide significantly reduced HbA1c and body weight versus placebo (substantial weight loss); the adverse events were similar in type to those of other GLP-1 agonists such as Semaglutide and Tirzepatide.

Three-receptor synergy: combines appetite suppression, insulin support and lipid mobilization — a broader mechanism than dual agonists.
Weight data: up to −24.2% at 48 weeks in the phase 2 trial; STEP 1 (Semaglutide) reported −14.9% at 68 weeks and SURMOUNT-1 (Tirzepatide) up to −20.9% at 72 weeks – separate trials, not a head-to-head comparison
Other endpoints: changes in liver fat, triglycerides and apoC-III (up to −40.6% and −38% respectively) — reported in clinical studies.
Contains a lipophilic concentrated segment, providing high stability and weekly administration.
Being studied in phase 3 programmes in obesity, type 2 diabetes and MASLD.

Retatrutide is an experimental triple agonist of the GLP-1, GIP and glucagon receptors, combining appetite suppression, increased energy expenditure and glycaemic regulation. In the phase 2 trial the highest-dose group showed a mean weight reduction of 24.2% at 48 weeks, together with marked changes in liver fat and metabolic markers. Phase 3 trials are ongoing; retatrutide is not an approved medicine, and the Peptology product is intended for laboratory research only.
